چکیده

BRCA1 mRNA and protein levels are regulated by the steroid hor monesestrogenand progesteronein humanbreastcancercells. BRCA1 mRNA and protein levels were significantly decreased in estrogen-de pleted MCF-7 and BT2OT cells and increased again after stimulation with @3-estradiol. The increase in BRCA1 expression upon stimulation with estrogen was not coordinated with the early induction of the estrogen dependentpS2 gene but closely paralleledthe delayedincreasein the S-phase dependent marker cydlin A. T47-D cells deprived of steroid hormones and subsequently stimulated with progesterone also showed a delayed increase in BRCA1 mRNA expression. However, no change in BRCA1 protein was detected in these cells. When considered together, the data suggest that steroid hormones may affect BRCA1 expression mdi racily by altering the proliferative status of the cells rather than acting directly on DNA sequences In the BRCA1 gene Itself.

Progesterone induces BRCA1 mRNA decrease, cell cycle alterations and apoptosis in the MCF7 breast cancer cell line.

BACKGROUND Inherited mutations of the BRCA1 gene are responsible for hereditary breast and ovarian cancer syndrome. However, little is known of how disruption of BRCA1 functions preferentially increases cancer risk in hormone-dependent organs. We aimed to study whether BRCA1 was regulated by progesterone in the MCF7 breast cancer cell line. MATERIALS AND METHODS MCF7 breast cancer cells were ...

Hormone-dependent regulation of BRCA1 in human breast cancer cells.

BRCA1 mRNA and protein levels are regulated by the steroid hormones estrogen and progesterone in human breast cancer cells. BRCA1 mRNA and protein levels were significantly decreased in estrogen-depleted MCF-7 and BT20T cells and increased again after stimulation with beta-estradiol. The increase in BRCA1 expression upon stimulation with estrogen was not coordinated with the early induction of ...

میزان بیان پروتئینBRCA1 در کارسینوم های مهاجم و درجای پستان و ارتباط آن با مارکر سلول های بنیادی سرطان پستان (CD44) و عوامل پیش آگهی تومور

Introduction : Breast cancer is the most common cancer and the second cause of cancer death among women. Despite recent developments in therapeutic tools about 25% of all the involved cases die annually. The clinical, molecular, and pathologic features of breast cancer in BRCA1 mutation carriers suggest that BRCA1 may function as a stem-cell regulator. The purpose of the current study was to ...

Synthesis Pyrano [2,3-c] Pyrazole-based compounds to induce apoptosis by reducing the expression of anti-apoptotic Bcl-2 protein in human breast cancer MCF-7 cells

Aim: Bcl-2 is a potential target for tumor treatment. The inhibition of the Bcl-2 production is research target of attract in the field of anti-cancer drug development. Recently, the assessment of antitumor activity appeared to be promising for pyrazole derivatives. Therefore, this study was designed to investigate the anti-cancer effects of novel pyrazole derivatives (HN1and HN2.). Material an...

Concerted Transcriptional Regulation by BRCA1 and COBRA1 in Breast Cancer Cells

Cofactor of BRCA1 (COBRA1) was first identified as a protein that binds to the breast cancer susceptibility gene product BRCA1. COBRA1 modulates estrogen-dependent and independent transcription and suppresses the growth of breast cancer cells. Its expression is significantly reduced in metastatic and recurrent breast cancer, pointing to a tumor suppressor function in breast cancer development. ...
